Dereplication Study on Glaucium aleppicum Boiss. in Jordan

چکیده
The secondary metabolite constituents of Glaucium aleppicum Boiss. et Hausskn. ex Boiass. (Papaveraceae) were investigated using an in-house developed Ultra Performance Liquid Chromatography-Photodiode Array-High Resolution Tandem Mass Spectrometry (UPLC-PDAHRMS-MS/MS) method. In this study the powerful resolution and short analysis time afforded by UPLC coupled to the outstanding mass accuracy of an Orbitrap mass spectrometer was used to develop a rapid and effective dereplication method to identify secondary metabolite constituents of G. aleppicum in crude extracts. In addition, HCD MS/MS and UV (photodiode array) spectra were used as an integral part of the strategy. This methodology was applied on a small amount of extract (~ 1 mg) of G. aleppicum to dereplicate twelve compounds. Five of which have been isolated previously from G. aleppicum: allocryptopine, protopine, corydine, isocorydine, and norchelidonine. Seven were new to the species, these include: dehydroglaucine, norisocorydine, chelerythrine, norsanguinarine, dihydrochelerythrine, corytuberine, and fumaflorine methyl ester. In addition to one more compound that was unidentified and thus considered new. keywords: Norisocorydine, Norchelidonine, Dihydrochelerythrine, Corytuberine, Norsanguinarine, Fumaflorine methyl ester, Chelerythrine, Dehydroglaucine, Glucium aleppicum, LC-MS.

متن کاملColchicinoids from Colchicum crocifolium Boiss.: a case study in dereplication strategies for (-)-colchicine and related analogues using LC-MS and LC-PDA techniques.
As a part of a project designed to investigate Colchicum species in Jordan, the chemical constituents of Colchicum crocifolium Boiss. (Colchicaceae) were investigated using LC-MS and LC-UV/Vis PDA. A decision tree for working with colchicinods has been developed by incorporating data from LC-UV/PDA and LC-MS. This dereplication strategy draws upon the UV/PDA spectra to classify compounds into o...

متن کاملIdentification and Quantification of the Main Active Anticancer Alkaloids from the Root of Glaucium flavum
Glaucium flavum is used in Algerian folk medicine to remove warts (benign tumors). Its local appellations are Cheqiq el-asfar and Qarn el-djedyane. We have recently reported the anti-tumoral activity of Glaucium flavum root alkaloid extract against human cancer cells, in vitro and in vivo. The principal identified alkaloid in the extract was protopine. This study aims to determine which compone...
